Natural Rapalogs Supplement

How do mTOR Inhibitors Work?

The mTOR inhibitor (mechanistic target of Rapamycin) is a threonine and serine protein kinase that was discovered over two decades ago. Since its discovery, it's become clear mTOR plays a role in governing cell proliferation and growth, making it an ideal target against conditions caused by cell proliferation.

How mTOR Inhibitors and Aging Are Connected

The acronym mTOR, which stands for mammalian target of Rapamycin, refers to an enzyme from the serine/threonine protein kinase family encoded by the mTOR gene. It is found in humans as well as worms, mice, flies and yeasts. It regulates growth, proliferation, motility, and survival of cells.

Cell senescence seems to be the driving force behind aging, both in terms of loss of cell-replication potential as well as mTOR signaling pathways inducing too much growth.

In fact, successfully inhibiting mTOR signaling pathways has been shown to affect the aging of worms, flies, yeasts and even mice – when accompanied by calorie restriction.

Benefits of Natural Rapalogs

Rapamycin is a molecule produced naturally by the mycelial bacterium Streptomyces hygroscopicus, which was discovered on Easter Island in 1975. It is an immunosuppressant drug used in orthodox medicine to prevent rejection following organ transplantation.

Rapamycin inhibits the mTOR signaling pathway and studies show it may contribute to extending the lifespan of mammals, even when it is taken in later life.

Other studies have identified potential benefits for age-related cognitive decline, specifically involving memory and learning capacity, and in particular, decreases in certain proteins that are often associated with neurodegenerative conditions. In pre-clinical trials, rapamycin analogs or "rapalogs," in particular inhibitors of the complex mTORC1, have shown measurable efficacy against certain cancers.

What are the Ingredients in Natural Rapalogs?

Rapalogs’ definition is synonymous with its function: an mTOR inhibitor, or mTOR inhibitor supplement. Our natural rapalogs mTOR supplement contains the following ingredients:

Curcumin

Meriva® is a patented curcumin formulation (derived from Curcuma longa) combined with soy phospholipids to increase its bioavailability beyond that of classic standardized curcumin extracts.

This key Curcuma ingredient has emerged as a natural mTOR inhibitor.

Green tea extract standardized to 70% EGCG

Studies have confirmed the inhibitory role of certain components of green tea (Camellia sinensis) on mTORC1 and mTORC2. Specifically, a study conducted by GlaxoSmithKline in Collegeville, Pennsylvania, showed that EGCg (epigallocatechin-3-gallate) acted as a double inhibitor, both of the P13K (phosphoinositide-3-kinase) pathway and the mTOR signaling pathway.

Resveratrol

Research has established that trans resveratrol, a polyphenol compound and calorie restriction mimetic, also inhibits the mTOR signaling pathway. Studies show that this pathway is activated by oxidized LDL-cholesterol lipoproteins in smooth muscle cells. Resveratrol can, however, block the phosphorylation and activation of the P13K and mTOR pathways. This activity is independent of resveratrol's antioxidant effect and its activation of AMPK and clearly identifies a new property for resveratrol that may add to this polyphenol's overall anti-atherogenic capacities. Another study reported that at relatively high doses, resveratrol inhibits mTOR and cell senescence while contributing to cell apoptosis.

Fisetin

This flavonoid, extracted from the small tree Rhus succedanea, sends an activating signal to anti-aging gene carrier cells, ensuring DNA and neuron protection, particularly during periods of oxidative stress.

Fisetin also has a stabilizing effect on resveratrol by preventing its metabolic breakdown and therefore supports its function of inhibiting the mTOR signaling pathway.

Salicin extracted from white willow

This highly active compound extracted from white willow bark gave rise to the isolation of the acetylsalicylic acid molecule (aspirin®) by the chemist Felix Hoffman.

Salicin possesses all the activity of aspirin®, including its mTOR inhibitory effect. Although not as fast-acting as aspirin®, its effect is less intrusive, and it has longer-lasting efficacy.

How to Take mTOR Inhibitor Supplements?

Even the most effective rapalogs will not work properly if you do not take them correctly. If you are taking rapalogs for aging, we recommend taking 3 capsules a day, always with a meal.

Note, however, that before you begin taking any supplement, you should research the side effects.

There is also a chance that Natural Rapalogs could interact negatively with other medicines. For example, Rapalogs should not be taken if you are allergic to aspirin. There are also some conditions that could prevent you from taking this supplement. If you are taking any other prescriptions or supplements, or if you have any preexisting health conditions, you will want to discuss this supplement with your healthcare provider.

It is unclear how rapalogs supplement may affect pregnant or breastfeeding mothers. For this reason, we recommend avoiding taking this supplement until more research is performed.
